The bookish unicorn
The studious librarian of the Golden Oak library and the heart of the game's main secret path, Twilight's Experiment. Her window is reached by flight, and her storyline ties into stealing and returning Trixie's book.
How to unlock: As a pegasus, fly to the library window and help Spike with a 50-bit train ticket, then return that night.
The speedster pegasus
Ponyville's fastest flyer, who lounges on a private cloud you can only reach with wings during the day. Her route is gated behind a short reflex race, so she rewards quick clicking.
How to unlock: Get wings from Trixie and fly up to her cloud during the day, then clear the race mini-game.
The party pony
The bubbly party pony from Sugarcube Corner and one of the easiest early routes. In the final 1.5 build her storyline closes with the "Goodbye Equestria" easter egg hidden in the box the next day.
How to unlock: Trigger her mini-game, then return the next day and click the box for the secret scene.
The fashionista
The refined dressmaker who runs the Carousel Boutique in town. Her path teaches Magic Attack B as a unicorn, then needs a return trip by flight at night, so it spans two forms.
How to unlock: Visit the Carousel Boutique, learn Magic Attack B, cut the upstairs window the next day, then return that night as a pegasus.
The apple farmer
The hard-working farmer of Sweet Apple Acres and the classic first target, because her route mixes two forms. You break the rock on her path as a unicorn, then buck apples as an earth pony.
How to unlock: Learn Magic Attack A, get a horn from Trixie, clear the rock at Sweet Apple Acres and buck the apples.
The shy animal lover
The gentle animal caretaker who lives in a cottage on the edge of the Everfree Forest. Her cottage stream matters for a later route: drinking from it first is what keeps you safe on the Zecora path.
How to unlock: Approach her gently at her cottage near the forest edge.
Ruler of the sun
The sun princess who rules from Canterlot Castle high above Ponyville. She is one of two royals you can reach, and her route needs wings and daylight to fly up to the castle.
How to unlock: Take the pegasus form, fly to the castle by day and enter from the left.
Ruler of the night
The night princess and the game's true endgame. She is the hardest character to reach because her fight is locked to night 3 and demands a pegasus plus several earlier scenes; beating her shield battle grants the alicorn form.
How to unlock: Endgame: on night 3, fly to the castle as a pegasus and win her shield battle to earn the alicorn form.
DJ Pon-3
Ponyville's turntablist, better known as DJ Pon-3. Her house sits past a record-shaped arrow, and clicking her window after the mini-game plays out the "Remixing Time" gallery scene.
How to unlock: Enter her house past the record arrow, then click her window after the mini-game for the Remixing Time scene.
The boastful magician
The boastful showmare who summons you into Ponyville and doubles as the game's hub. She hands out the horn and wings that gate the other forms, and her own book is the key item for Twilight's Experiment.
How to unlock: Find her in town - she also hands you the horn you need for other paths. As a unicorn at night, break her shield to take her book.
The zebra alchemist
The mysterious zebra alchemist who keeps a hut deep in the Everfree Forest. Reaching her means solving the forest maze - but only after drinking from Fluttershy's stream, or the trip poisons you.
How to unlock: Drink from Fluttershy's stream first, head into the Everfree Forest, then solve the maze to reach her hut.
The changeling queen
The changeling queen, and the Everfree maze's hidden twist. Run the exact same forest route but skip Fluttershy's stream, and the pony you thought was Zecora is revealed to be Chrysalis in disguise.
How to unlock: Do the Everfree maze but do NOT drink Fluttershy's stream - "Zecora" is revealed as the disguised Chrysalis.
The clumsy mailmare
The lovable wall-eyed mailmare, and the newest face in the roster: her #SaveDerpy mailbox mini-game was added in the final 1.5 build. Free her from the mailbox to open her scene.
How to unlock: Click the #SaveDerpy mailbox three times and choose Yes to free her (mini-game added in 1.5).

Princess Luna. Her fight is locked to the third night and needs the pegasus form plus several earlier scenes done first, so she is effectively the Banned from Equestria endgame. Winning her shield battle grants the alicorn form, the final reward.
There are two royal sisters you can reach in Banned from Equestria. Celestia needs the pegasus form during the day - fly up to Canterlot Castle and enter from the left. Luna is the night-3 endgame: fly to the castle as a pegasus and win her shield battle.
Yes - Queen Chrysalis is in Banned from Equestria as a hidden twist on the Zecora route. Run the Everfree Forest maze but skip drinking from Fluttershy's stream first, and the pony posing as Zecora is revealed to be Chrysalis in disguise.
The final 1.5 build has the largest roster in the series: the Mane 6 (Twilight, Rainbow Dash, Pinkie Pie, Rarity, Applejack and Fluttershy), Princess Celestia and Luna, plus Vinyl Scratch, Trixie, Zecora, Queen Chrysalis and Derpy, whose #SaveDerpy mini-game was added in 1.5.
